چکیده

The BAX gene is characterized by 5 protein coding transcripts (alpha/psi, beta, delta, epsilon, sigma). Baxbeta encodes the longest isoform (891 bp) of the gene. BAX-alpha/Bax-psi variant is 888 bp in length and codes for a protein isoform that possesses a shorter and different C terminus, as compared with the isoform BAX-beta. The third variant (BAX-delta), which is 741 bp in length, lacks exon 3, whereas it retains the functionally critical C-terminal membrane anchorage region, as well as the BCL2 homology 1 and 2 (BH1 and BH2) domains, although it has a shorter and different C terminus, in comparison with BAX-beta. The fourth identified variant of BAX, which is designated as BAX-epsilon, is 986 bp in length because it contains an extra fragment within the coding region, as well as a distinct 3' coding region and 3' UTR, resulting in a distinct BAX isoform with a shorter and distinct C terminus, as compared with BAX-beta. The fifth identified variant of BAX, BAX-sigma, is 849 bp in length and has also a shorter and different C terminus, when compared with the isoform beta. Pseudogene

Expression of Apoptosis Related and Proliferative Proteins in Malignant Lympho-Proliferative Disorders

Background & objective: The current study aimed to perform an immunohistochemical analysis of patterns of apoptotic and cell proliferative related protein expression in different histological grades and immune phenotypes of malignant lymphomas and other lymphoproliferative disorders Methods:This observational study was carried o...

Bcl-2, Bcl-X, Bax, and Bak expression in short- and long-lived patients with diffuse large B-cell lymphomas.

Long-term cure is now possible for approximately 50% of all patients with aggressive non-Hodgkin's lymphoma (NHL). Apoptosis-related proteins play an important role in the chemosensitivity or chemoresistance of tumors. We examined the role of Bcl-2 family proteins in aggressive NHL. We retrospectively selected two groups of patients by clinical outcome: 24 patients with chemoresponsive disease ...

Apoptosis and expression of Bax, Bcl-x, and Bcl-2 apoptotic regulatory proteins in colorectal carcinomas, and association with p53 genotype/phenotype.

AIMS Spontaneous apoptosis and expression of the apoptotic regulatory proteins Bax, Bcl-x, and Bcl-2 were investigated in 50 colorectal carcinomas. The p53 genotypes/phenotypes and BAX genotypes were also determined, and possible associations of these with apoptosis and/or with expression of the different apoptotic regulatory proteins were studied. METHODS Terminal deoxynucleotidyl transferas...

PUMA promotes Bax translocation by both directly interacting with Bax and by competitive binding to Bcl-X L during UV-induced apoptosis.

Cell apoptosis induced by UV irradiation is a highly complex process in which different molecular signaling pathways are involved. p53 up-regulated modulator of apoptosis (PUMA) has been proposed as an important regulator in UV irradiation-induced apoptosis. However, the molecular mechanism through which PUMA regulates apoptosis, especially how PUMA activates Bcl-2-associated X protein (Bax) in...

The great migration of Bax and Bak

The Bcl-2 proteins Bcl-2-associated X protein (Bax) and Bcl-2 antagonist killer 1 (Bak) can commit cells to apoptosis. Retrotranslocation of Bax from the mitochondria into the cytosol is essential for cell survival. Recently, we reported that Bak is also present in the cytosol of healthy cells and that Bax and Bak are regulated by the same retrotranslocation process.

Epigenetic determinants of resistance to etoposide regulation of Bcl-X(L) and Bax by tumor microenvironmental factors.

BACKGROUND Epigenetic factors (i.e., alterations of gene activity not involving mutations), as well as genetic changes in surviving cancer cells, may play an important role in drug resistance following cancer chemotherapy-a common cause of tumor relapse. Bcl-2 family proteins are central to the regulation of apoptotic cell death and modulate drug sensitivity. We investigated how survival signal...
